Single-molecule FRET Imaging at St. Jude

In the Single-Molecule Imaging Center at St. Jude Children’s Research Hospital, scientists engineer and employ bright fluorescent probes to investigate individual molecules and their interactions. To obtain accurate images, highly advanced, custom-built microscopes are used. Multiple lasers are co-aligned and then focused by a series of high-power lenses and other specialized optics, to a very small volume, where one molecule at a time can be observed. This approach, called single-molecule FRET (fluorescence resonance energy transfer) imaging, allows scientists to track the movements of each part of functioning single molecules in real time. Characteristic features of the FRET signals originating from individual molecules can reveal both how they correctly function and how they malfunction. Discoveries could lead to new treatment options and more effective clinical care for patients.
