It can grow rapidly on minimal medium that contains a carbon compound such as glucose (which serves both as a carbon source and an energy source) and salts which supply nitrogen, phosphorus, and trace metals. E. coli grows more rapidly,however, on a rich medium that provides the cells with amino acids, nucleotide precursors,vitamins, and other metabolites that the cell would otherwise have to synthesize. Most strains of E. coli can be stored for years in stab vials, or indefinitely if frozen at −70°C. It is prudent to check the genetic markers of a strain revived from storage. Effective storage means that the organism is being maintained in a viable state without contamination and changes in genotypic or phenotypic characteristics. The organism must be easily restored to the same condition it was in prior to preservation. Several methods are available for the preservations of the microorganisms but there are two criteria for selecting a preservation method for a given culture. They are: i) period of preservation desired and ii) nature of culture to be preserved. For different physiological groups of bacteria, specific maintenance requirements exist.
