#This video demonstrate a spectrum measurement using UV-1900i standalone operation. You can also see the how survey scan speed of UV-1900i, which is one of the key features of the instrument being used for spectrum measurement. UV-Vis spectroscopy is an analytical technique that measures light absorption of a sample across the ultraviolet and visible range of the electromagnetic spectrum. This can be done using an instrument known as a spectrophotometer, such as the UV 1900i, which provides the data in the form of an absorption spectrum. This displays how much light is able to pass through the sample and into the detector of the spectrophotometer. The absorption spectrum is normally presented in the form of graph, where the y axis represents absorbance, and the x axis represents wavelength Peaks in the spectrum, known as absorption peak, indicates the wavelength of light that is absorbed by the sample. Now, lets look at how the UV 1900i is used to measure the spectrum of a liquid sample. From the mode menu screen, we will select spectrum. Set the measurement parameter required for the analysis, such as wavelength scan range and photometric recording range. Then, we are going to set the wavelength scan speed. For this example, we are going to select survey which can perform measurement with speed up to 29,000 nm/min. This means the spectrum scan can be done within a few seconds. Now, we are going to perform baseline correction. Place the cuvette containing blank reagent into both sample and reference cells of the sample compartment. Select baseline correction from the screen. After baseline correction is finished, replace the blank reagent with the sample solution for the sample cell. Now, select start to perform the spectrum measurement. The spectrum measured is being displayed on the measurement screen. From the spectrum, we can see some absorbance peaks. To determine the absorbance peak wavelength, first, select operations. Followed by peak pick. Peak detected would be displayed by the small white indicators.
