EMERGE Episode 10 : Trapped ion mobility-resolved (phospho)proteomics of clinical samples

Dr Florian Meier presents on quantitative mass spectrometry (MS)-based proteomics of clinical specimens offers unique opportunities to advance our understanding of human health and disease. Conventional workflows digest complex protein extracts with proteolytic enzymes and analyze the resulting peptide mixture via liquid chromatography – MS. Recently, advances in efficiency and ease of implementation have rekindled the interest in ion mobility spectrometry as an additional dimension of separation. Trapped ion mobility spectrometry (TIMS) features a particularly compact design and can be readily integrated with rapid quadrupole time-of-flight mass spectrometers. We have recently demonstrated that synchronizing the release of ions from the TIMS device with downstream precursor selection multiplies the speed and sensitivity of peptide sequencing in a novel scan mode termed PASEF. Making use of the correlation between ion mobility and mass, the PASEF principle also boosts the efficiency of data-independent acquisition (dia-PASEF). This presentation will showcase applications of dia-PASEF to analyze the proteome and phosphoproteome of clinical samples with high throughput and high sensitivity.
