It is currently accepted that environmental and genetic factors act together to induce this pathology. It is therefore referred to as a complex pathology. To date, five constitutional abnormalities of the coagulation cascade are known to promote VTE: three deficiencies in coagulation inhibitors, antithrombin (AT), protein C (PC), protein S (PS) and two genetic variants’ located respectively on the genes for factors V and II, factor V Leiden and G20210A. These five abnormalities constitute the classic 'thrombophilia check-up' (constitutional) recommended by learned societies.
