TSS: PacBio presents the Revio and Onso systems for HiFi and sequencing by binding (SBB) sequencing

Seminar Abstract: Today’s most popular DNA sequencing platforms produce reads that are 300 bp long with base accuracy of around 99.9% (Q30). ‪@PacificBiosciences‬ recently two new platforms – the Revio long-read sequencing system and Onso short-read sequencing system – that extend these attributes to answer new biological questions. The Revio system features PacBio HiFi sequencing, which produces reads with Q30 accuracy and 15,000 – 20,000 bp in length. These long, accurate reads provide the most comprehensive view of genomes with the ability to access new classes of variation (structural variation and tandem repeats) and new regions of the genome like segmental duplications and centromeres. Further, HiFi reads add the ability to phase variants into haplotypes and directly detect DNA methylation in standard sequencing libraries, without any special sample treatment. HiFi reads were key to assembling the first truly complete assembly of a human genome, the telomere-to-telomere CHM13 reference, and characterizing its epigenome. The Onso system features PacBio sequencing-by-binding (SBB), which produces 200-300 bp reads with base accuracy exceeding 99.99% (Q40). This breakthrough accuracy reveals rare events – like somatic variant in circulating tumor DNA – that are below the detection limit of Q30 sequencing platforms. The near-perfect accuracy of Onso identifies rare events in single molecules and eliminates the need to use approaches like unique molecular identifiers to extend accuracy. This in turn reduces the required sequencing coverage, as redundant oversampling is not necessary.
